Abstract
Design and synthesis of novel spirocyclic compounds is a challenging task in organic and medicinal chemistry because of the generation of quaternary center, which holds central chirality or an axial chirality. This chapter includes various strategies that utilize metathesis for the construction of spirocycles in combination withother well-known name reactions, such as Claisen rearrangement, Grignard addition, Fischer indolization, Suzuki Miyaura, cross-coupling, and retro Diels Alder reaction. The catalysts used to realize the metathesis sequence for the synthesis of spirocycles are Grubbs first-generation catalyst, Grubbs second-generation catalyst, Hoveyda Grubbs first-generation catalyst, and Hoveyda Grubbs second-generation catalyst. The chapter provides the information on the formation of aza-spirocycles, and the formation of oxa-spirocycles.
